Protein Assays

Uptima BC & Micro BC Assays

Uptima BC & Micro BC Assays are cost effective formulations designed to match leading brand performance.

They are based upon the interaction of bicinchoninic acid with a Cu1+ complex that is formed when proteins interact with Cu2+.

These assays demonstrate excellent linearity compared to Coomassie based assays and are compatible with a range of detergents and other substances (see instructions).

BC Assays display a working range from 5 -to- 2000ug/ml (protocol dependant) whilst the Micro BC Assay's range is from 1 -to- 200ug/ml.

Uptima CooAssay's

The use of a Coomassie dye for total protein determination was cited by Dr Marion Bradford in the mid 70's and modified ready to use formulations are widely adopted in laboratories performing biological processes.

Uptima CooAssay's are cost effective formulations designed to match leading brand performance. CooAssay Standard is a classic formulation whilst CooAssay Max is an enhanced formulation that improves curve linearity. Both Assays require no pre-preparation and are ready to assay in seconds.

Reagents have been independantly tested to verify their capability.

General Considerations

Which Protein Assay?

This is generally driven by your historical laboratory experience. The basic rule of thumb is that a CooAssay will provide the fastest, easiest means to assay protein and works well in reducing substances. BC Assays are the option if you want improved curve linearity and sensitivity and are working in the presence of detergents. Most labs will have both types available in their laboratories

Which protein standard?

A highly purified version of the protein of interest is always preferable. This is not always possible/necessary and in these situations Bovine Serum Albumin (BSA) is a good option. If you are using antibodies then go for Bovine Gamma Globulin (BGG). Construct a standard curve each time an assay is performed.

Removing interfering substances.

Dialysis, gel filtration or precipitate proteins with cold acetone or trichloroacetic acid.

If protein concentration is high, dilute the sample to a level which will not cause interference.

What wavelength should I use?

562nm is optimal. You can measure the purple colour between 540nm and 590nm however, this will result in a lower slope for the standard curve and may increase the minimum detection level.

What interferes with the BC Assay?
